Homology modelling and analysis of structure predictions of human tumour necrosis factor ligand superfamily member 8

Abstract Background Tumour necrosis factor ligand superfamily member 8 (TNFL8) is a cytokine that plays vital roles in immune activations and inflammatory responses through its interaction with the tumour member. Despite multiple studies on involvement of receptor response, there limited information molecular characterization structural elucidation cytokine. Considering significance cytokine, three-dimensional structure TNFL8 model was generated by homology modelling Iterative Threading ASSEmbly Refinement (I-TASSER) server validated PROCHECK Qualitative Model Energy Analysis (QMEAN) servers. Results The predicted has 90.00% residues most favoured region Ramachandran plot while QMEAN value gives − 3.06. sequence alignment between template (1XU2) reveal similar active site such as ILE-142, THR-175, GLU-178, PHE-228 could be involved binding pocket formation. However, docking 3D eight phytochemicals from extract Xylopia aethiopica (Dunal) A. Rich revealed phytochemical bound to two different sites which regions essential for inhibition. More so, analysis showed have good affinity ellagic acid showing highest energy 6.58 ± 0.18 kcal/mol. Conclusion proposed may shed light mechanisms provide insights into identification potential targets development novel compound regulation functional activities
